Tesamorelin is a synthetic 44-amino-acid GHRH analogue with an N-terminal trans-3-hexenoic acid modification that protects against DPP-4 cleavage. It is used as a full-length GHRH reference compound in pituitary receptor studies and in models of visceral adipose tissue biology.

Tesamorelin is a 44-amino-acid synthetic analogue of the entire human GHRH sequence — distinct from Sermorelin (29-AA fragment) and CJC-1295 No DAC (modified fragment) — with a trans-3-hexenoic acid modification at the N-terminus that confers resistance to DPP-4 enzymatic cleavage while preserving the full-length GHRH structure. It activates the GHRH receptor on anterior pituitary somatotroph cells through Gs protein coupling, cAMP elevation and calcium-dependent growth hormone secretion. As a full-length GHRH analogue with enhanced stability, it produces growth hormone release with pharmacokinetics intermediate between endogenous GHRH (minutes half-life) and structurally modified long-acting analogues. Tesamorelin has been characterised as a reference compound for studying GHRH receptor activation using the complete native peptide sequence.

The most extensively characterised GHRH analogue in clinical research, with a formal regulatory history. FDA approval: approved in 2010 for HIV-associated lipodystrophy (Egrifta, Theratechnologies) — the only GHRH analogue to receive regulatory approval for a clinical indication; a second approval (Egrifta SV) followed in 2019. Pivotal trials: two randomised controlled Phase 3 trials (IRISS studies) demonstrated reduction of visceral adipose tissue in HIV-positive individuals on antiretroviral therapy. Mechanistic characterisation: pituitary responsiveness and GH pulsatility studies established the pharmacodynamic profile for the full-length analogue class and served as a comparator for fragment and modified-fragment variants. Additional research examined GH-mediated effects on body composition, lipid metabolism and cognitive measures in specific populations. Note: GHRH analogues are listed by anti-doping authorities; FAB Research supplies this compound for laboratory research only, not for any clinical purpose.

  • 5 mg per application (one metered dose)
  • 4 doses per pen (20 mg total)
  • Full-length 44-amino-acid GHRH analogue — distinct from Sermorelin (29 AA) and CJC-1295 No DAC (modified fragment)
  • Pre-filled sterile solution — no reconstitution step required

Storage: Store refrigerated at 2–8 °C in the original sealed box. Do not freeze. Protect from light.

  • For in-vitro laboratory and preclinical research only — not for human or animal administration
  • Tesamorelin has a formal FDA approval history (Egrifta, for HIV-associated lipodystrophy) — provides a more complete clinical safety reference than most experimental secretagogues
  • Glucose metabolism effects are documented in the Egrifta prescribing information — studies involving glucose-sensitive endpoints must account for this
  • DPP-4 resistance (trans-3-hexenoic acid N-terminal modification): the full GHRH sequence with stabilisation distinguishes it from fragment-based analogues

  • From Egrifta (FDA-approved) prescribing information: injection site reactions (11.5%), peripheral oedema (6.0%), arthralgia (6.0%), myalgia
  • Glucose metabolism effects: blood glucose elevation documented in Egrifta label — monitor in glucose-sensitive assay designs
  • Headache, dizziness, flushing in clinical studies
  • Fluid retention consistent with GH axis activation
  • One of the best-characterised clinical safety profiles among research secretagogues due to its prior regulatory approval history

Effects listed are observations from published research literature and are not predictions for any specific use. All FAB Research products are for in-vitro laboratory research — not for human or animal administration.
