Cagrilintide is a lipidated amylin analogue engineered for extended receptor residence time at the amylin and calcitonin receptors. In preclinical literature it is studied both alone and alongside GLP-1 agonists to model additive effects on satiety signalling and energy balance. Supplied as a sterile pre-filled metered research pen.
Cagrilintide is a lipidated long-acting amylin analogue that activates amylin receptors — heteromeric complexes of the calcitonin receptor (CTR) with receptor activity-modifying proteins RAMP1, RAMP2 and RAMP3 (forming AMY1, AMY2 and AMY3 respectively). This is mechanistically distinct from GLP-1 receptor-based compounds; the two receptor systems are investigated for complementary, additive effects on satiety signalling and metabolic regulation. Amylin receptor signalling engages cAMP and intracellular calcium pathways. A C20 fatty acid modification enables albumin binding and extends half-life substantially beyond that of native amylin. Research has examined cagrilintide both as a standalone amylin analogue and in fixed-ratio combination with semaglutide (CagriSema) to probe additive effects through independent receptor pathways.
A developing clinical literature focused on pharmacokinetics, safety and metabolic endpoints. Phase 2 CagriSema trials: combination studies with semaglutide investigating additive effects through GLP-1 and amylin receptor co-activation, with published metabolic and body composition findings. Standalone amylin pharmacology: preclinical and early clinical characterisation of receptor binding, downstream signalling and dose-response relationships for the lipidated analogue. Satiety signalling: investigation of central mechanisms including area postrema, nucleus tractus solitarius and hypothalamic circuits through which amylin receptor activation influences appetite regulation. Evidence status: investigational compound currently in Phase 3 development (REDEFINE programme); data reflect ongoing industry-sponsored research with no regulatory approvals at time of publication.
- 2.5 mg per application (one metered dose)
- 4 doses per pen (10 mg total)
- Pre-filled sterile solution — no reconstitution step required
- Amylin / calcitonin receptor agonist — mechanistically distinct from GLP-1 agonists; suitable for complementary co-exposure designs
Storage: Store refrigerated at 2–8 °C in the original sealed box. Do not freeze. Protect from light.
- For in-vitro laboratory and preclinical research only — not for human or animal administration
- Acts via amylin receptor (CTR + RAMP heteromers) — distinct pathway from GLP-1 / GIP receptor agonists; complementary rather than overlapping
- Investigational compound (Novo Nordisk REDEFINE Phase 3); independent published literature is limited
- For co-exposure designs with semaglutide (CagriSema model): each compound should be sourced and dosed independently for reproducibility
- Phase 2 CagriSema data: nausea, vomiting, diarrhoea, constipation — amylin receptor class effects, qualitatively similar to GLP-1 agonism
- Injection site reactions
- Decreased appetite
- Evidence base is industry-sponsored Phase 2–3 data; independent characterisation is limited
Effects listed are observations from published research literature and are not predictions for any specific use. All FAB Research products are for in-vitro laboratory research — not for human or animal administration.
