5-Amino-1MQ (5-amino-1-methylquinolinium) is an experimental small-molecule NNMT inhibitor. It acts by inhibiting nicotinamide N-methyltransferase (NNMT), an enzyme that converts nicotinamide into 1-methylnicotinamide using SAM as a methyl-group donor. Because nicotinamide feeds the NAD+ salvage pathway, NNMT activity connects nicotinamide methylation with NAD+ availability. Laboratory and animal studies have investigated NNMT inhibition in relation to adipocyte biology, lipid metabolism and cellular energy regulation. FAB Research supplies 5-Amino-1MQ in a 100mg pre-filled metered research pen for laboratory investigation.
By blocking NNMT-catalysed methylation of nicotinamide, 5-Amino-1MQ potentially increases nicotinamide availability for the NAD+ salvage pathway — a mechanism investigated in adipocyte culture experiments. The NNMT reaction also consumes SAM, so inhibition alters the cellular SAM-to-SAH ratio, linking the compound to methylation-associated signalling. Its membrane permeability allows intracellular NNMT access. These mechanisms are distinct from NAD+ precursors (NMN, NR) and from GLP-1 receptor-mediated metabolic signalling.
A 2018 study in Biochemical Pharmacology investigated NNMT inhibitors in diet-induced obese mice, reporting reductions in body weight and adipose tissue mass — animal findings not interpretable as clinical evidence. Adipocyte culture experiments reported changes in 1-MNA, NAD+ and SAM concentrations. Small-molecule pharmacology research characterised enzyme selectivity and structure-activity relationships. The compound remains preclinical: safety, effectiveness and pharmacological behaviour in humans have not been established.
- 10 mg per application (one metered dose)
- 10 doses per pen (100 mg total)
- Small-molecule NNMT inhibitor (not a peptide) — included for use alongside peptide compounds in NAD⁺ salvage and metabolic research
- Pre-filled sterile solution — no reconstitution step required
Storage: Store refrigerated at 2–8 °C in the original sealed box. Do not freeze. Protect from light.
- For in-vitro laboratory and preclinical research only — not for human or animal administration
- NNMT inhibitor mechanism is distinct from NAD⁺ precursors (NMN, NR) and GLP-1 receptor agonists — appropriate controls for the specific pathway (nicotinamide methylation, SAM:SAH ratio) required
- Only preclinical animal and cell-culture data available — no human clinical trial data; safety in humans is unknown
- Membrane-permeable small molecule: intracellular access to NNMT is the mechanism — confirm intracellular concentration estimates for in-vitro assay designs
- Only preclinical data available — no human adverse effect profile established
- 2018 Biochemical Pharmacology mouse study (Neelakantan et al.): generally well tolerated at studied doses in diet-induced obese mice
- No human safety studies published; clinical adverse effect profile is entirely unknown
- Cell-culture toxicity: characterise concentration-dependent effects in target cell lines before designing major experiments
Effects listed are observations from published research literature and are not predictions for any specific use. All FAB Research products are for in-vitro laboratory research — not for human or animal administration.
